General description

2′-Deoxycytidine 5′-triphosphate (dCTP) is synthesized either by de novo pathway or from the multiple phosphorylation steps of cytidine. It is deaminated to deoxyuridine monophosphate (dUMP) which is convertible to deoxythymidine triphosphate (dTTP).

Application

2′-Deoxycytidine 5′-triphosphate disodium salt has been used:
  • in polymerase chain reaction mix for the amplification of Alternaria (AM) toxin DNA[1]
  • to spike oligonucleotide sample for capillary electrophoresis (CE) analysis[2]
  • as a component in adenosine 5′-monophosphate (AMP) luciferase assay[3]

2′-Deoxycytidine 5′-triphosphate disodium salt is one of the nucleoside triphosphates used for DNA polymerase driven reactions such as polymerase chain reaction, DNA sequencing and other molecular biology methods.

Biochem/physiol Actions

2′-Deoxycytidine 5′-triphosphate (dCTP) is used to synthesize DNA by DNA polymerases or reverse transcriptases. dCTP is an allosteric regulator of deoxycytidylate (dCMP) deaminase.[4] It is used in studies of nucleotide pools, nucleotide selectivity and enzyme specificity studies, such as the preferred incorporation of dCTP by Rev1, a γ-family DNA polymerase.[5]

A luciferase-based method was developed for measurement of 5'-adenylylsulfate (APS) reductase (APR), an enzyme of the reductive sulfate assimilation pathway in prokaryotes and plants. APR catalyzes the two-electron reduction of APS and forms sulfite and adenosine 5'-monophospahate (AMP). The luciferase-based
