Beschreibung

General description

Ein synthetisches, zellpermeables Actinobacteria-Pyrimidotriazindion, das die β-Catenin-Tcf4-Wechselwirkung selektiv unterbricht (IC₅₀ = 0,8 µmol/l in zellfreien Bindungsassays) und die durch β-Catenin regulierte zelluläre Transkriptionsaktivität hemmt (IC₅₀ = 0,3 µmol/l in HCT116-basierten Reporter-Assays). PKF118–310 kehrt die durch β-Catenin-, jedoch nicht die durch Siamois-mRNA eingeleitete Bildung der ventralen Körperachse in Xenopus-Embryos um, während eine direkte Injektion (1 mg/kg/0,5 Wochen) in die Tumorstelle von Mäusen mit HepG2-Xenotransplantat die Anzahl der apoptotischen Zellen steigert und die Expression von β-Catenin-Zielgenen (Cyclin D1, c-Myc, Survivin) in der Tumormasse verringert, was zu einer wirksamen Wachstumsverzögerung des Tumors in vivo führt (um ~60 % nach einer vierwöchigen Behandlungsdauer)
