Beschreibung

General description

Ein zellpermeables Dihydrothiopyranopyrimidinol, das TNKS1/PARP5a und TNKS2/PARP5b mit hoher Affinität bindet (K_d = 99 bzw. 93 nM) und deren PARsylierungsaktivität (Poly-ADP-Ribosylierung) stark hemmt (IC₅₀ = 11 bzw. 4 nM), während es eine viel geringere Aktivität gegen PARP1 und PARP2 aufweist (IC₅₀ = 2,194 bzw. 0,114 µM). Die Knockdown-Funktion von TNKS, nicht aber von PARP1/2, durch siRNA oder XAV939-Behandlung unterdrückt die zelluläre Axin1/2-PAR-Sylierung und Ubiquitinierung/proteasomalen Abbau, was zu Axin-Aufbau, β-Catenin-Zerstörung und Blockierung der Wnt-Signalübertragung führt. XAV939 hemmt nachweislich das Wachstum von β-catenin-abhängigen DLD-1-Zellen, aber nicht das von β-catenin-unabhängigen RKO-Zellen (um >95 % vs. keine Wirkung bei 3,3 µM) in vitro und verhindert die Regeneration der Zebrafisch-Schwanzflosse (um ~70 % bei 7 Tage Post-Amputation, 5 µM) nach chirurgischer Entfernung in vivo.
