Application

Carboanhydrase aus Rindererythrozyten wird zur Entwicklung von Kohlenstoffdioxid-Abscheidungssystemen und zur Erforschung verschiedener Aufreinigungstechniken verwendet . Darüber hinaus wird Carboanhydrase zur Untersuchung der Regulierung des Säure-Basen-Haushalts bei Fischen und zur Untersuchung des Carboanhydrase-II-Mangelsyndroms eingesetzt .

Biochem/physiol Actions

Carboanhydrase ist ein zinkhaltiges Enzym, das die reversible Umwandlung von Kohlendioxid in Bicarbonat katalysiert. Eines seiner physiologischen Hauptaufgaben ist die Beibehaltung des Säure-Basen-Gleichgewichts im Blut und anderen Geweben. Ein Mangel an Carboanhydrase führt zum Carboanhydrase-II-Mangelsyndrom, einer autosomal-rezessiven Erkrankung, die Osteopetrose, renale tubuläre Azidose und Verkalkungen im Gehirn verursacht .

Eine Wilbur-Anderson (W-A-)Einheit bewirkt eine Senkung des pH-Werts von 0,02 M Trizma-Puffer von 8,3 auf 6,3 pro Minute bei 0 °C. (Eine W-A-Einheit entspricht im Wesentlichen einer Roughton-Booth-Einheit.)

    1. The activity of this enzyme is established using a 0.02 M Tris buffer with a starting pH of 8.3 at 0°C. One unit will cause the pH of a 0.02 M Trizma buffer to drop from 8.3 to 6.3 per min at 0°C. However, the optimum pH and temperature is highly dependent on the specific application.
