Beschreibung

General description

Das fluorometrische Caspase-3-Assay Kit basiert auf der Hydrolyse von Acetyl-Asp-Glu-Val-Asp-7-amido-4-methylcumarin (Ac-DEVD-AMC) durch Caspase 3, durch die fluoreszentes 7-amino-4-methylcumarin (AMC) freigesetzt wird. Das Anregungsmaximum von AMC liegt bei 360 nm und das Emissionsmaximum bei 460 nm. Die Konzentration des freigesetzten AMC kann aus einer mit AMS-Standards erstellten Kalibrationskurve berechnet werden (AMC-Standard ist im Kit enthalten).

Application

Das fluorometrische Caspase 3-Assay-Kit ist für den Nachweis und die Messung der Caspase 3-Aktivität in Zellextrakten oder aufgereinigten Caspase 3-Präparationen vorgesehen.

Eine Einheit Caspase 3 spaltet 1,0 μmol Ac-DEVD pro Minute bei pH 7,4 und 25 °C.
