General description
Epitopmarkierungen bieten ein Verfahren zur Lokalisierung von Genprodukten in einer Vielzahl von Zelltypen, zur Untersuchung der Topologie von Proteinen und Proteinkomplexen, zur Identifizierung von assoziierten Proteinen und zur Charakterisierung von neu identifizierten, in geringer Anzahl vorkommenden oder nur schwach immunogenen Proteinen, wenn keine proteinspezifischen Antikörper verfügbar sind. Die Markierung mit der FLAG® Peptidsequenz kann am N-Terminus, am N-Terminus mit vorausgehendem Methioninrest, am C-Terminus oder an internen Positionen des Zielproteins vorgenommen werden. FLAG kann auch gemeinsam mit anderen Markierungen platziert werden. Die geringe Größe der FLAG-Markierung oder -Sequenz und ihre hohe Hydrophilie verringern oft die Möglichkeit von Störungen der Proteinexpression, proteolytischen Reifung, Antigenität und Funktion. Die N-terminale FLAG® Peptidsequenz enthält eine einzigartige Enterokinase-Spaltstelle, wodurch sie vollständig von den aufgereinigten Fusionsproteinen entfernt werden kann. Forschungsergebnisse weisen die durch Cu2+-Ionen katalysierte Spaltung des C-terminalen FLAG® Peptids von einem Fusionsprotein nach. Ein Sequenzmotiv mit fünf von acht Aminosäureresten, die mit dem FLAG® Peptid identisch sind, kommt in Mg2+-abhängigen Protein-b-Phosphatasen von Ratten und Mäusen sowie in humanen und bovinen Enzymen vor. Dieser ANTI-FLAG® Antikörper vom Kaninchen wird aus dem Hybridom SIG1-25 erzeugt. Das Hybridom wird aus der Fusion von Myelomzellen und Splenozyten von Kaninchen gewonnen, die mit der an KLH konjugierten FLAG® Peptidsequenz immunisiert wurden.

The main application for Product F2555 rabbit monoclonal is for immunoblotting after immunoprecipitation with M2, which is a mouse antibody. This enables using a secondary antibody (anti-rabbit) that will not bind the M2 mouse heavy (50 kD) and light chains (25 kD).
