The initial starting material used to prepare this product is citrated to prevent clotting. The whole blood is centrifuged and the plasma is separated. Calcium is then added to the plasma fraction to initiate clot formation. The resulting supernatant is then centrifuged to remove cellular debris.

Anwendungsgebiete von Humanserum: Als Bestandteil von phosphatgepufferter Kochsalzlösung mit Tween®-Detergens (PBST) im Bridging-ELISA (Enzyme-linked Immunosorbent Assay), als biologische Scheinprobe zur Beurteilung der Leistung des papierbasierten ELISA-Assays mit komplexen biologischen Proben, als Verdünnungsmittel zur Erstellung einer Verdünnungsreihe der Baseline-Probe für die Erstellung einer Standardkurve für NISTmAb als Zusatz zu Serum, sowie für die Multiples Myelom(MM)-Patientenprobe MM2232
Humanes AB-Serum wird für folgende Anwendungen eingesetzt: Gewebebearbeitung; Transplantations- und Zelltherapieanwendungen zur Expansion mesenchymaler Stammzellen aus adipösem Gewebe oder mesenchymaler Stromazellen aus humanem Knochenmark; Gewinnung und Transplantation limbaler epithelialer Stammzelltransplantate, Ex-vivo-Expansion von NK-Zellen aus peripherem Blut in Expansionsmedium für hematopoetische Stammzellen; Verbesserung der Kulturmethoden für humane Inselzellen vor der Transplantation.
Biochem/physiol Actions
Humanes AB-Serum wird in der Regel gewonnen, indem Vollblut auf natürliche Weise gerinnen kann, was als Off-The-Clot- bzw. OTC-Serum bezeichnet wird. Humanserum aus geronnenem Vollblut wird bevorzugt, da es in der Regel höhere Konzentrationen und ein vielfältigeres Spektrum von Wachstumsfaktoren für ein optimales Zellwachstum enthält. Humanserum von Spendern der Blutgruppe AB enthält keine spezifischen Antikörper gegen die Blutgruppenantigene A und B. Dadurch ist es ein universelles Spenderserum und für verschiedene Zellkulturtypen geeignet, einschließlich humaner Stammzelllinien.
Packaging
Die Verpackung kann sich aufgrund einer Knappheit der Flaschen, die zum Verpacken dieses Produkts verwendet werden, bei verschiedenen Chargen unterscheiden. Das Vorhandensein oder Fehlen von Schrumpffolie um den Flaschendeckel beeinträchtigt die Integrität/Sterilität des Produkts nicht.

Is there a significant difference in composition between different source batches (SLCQ2795 and 0000523739) of the same H4522-100ML serum? I used it to simulate human serum for detecting the EC50 values of compounds.

This serum product is pooled from multiple donors. Typical pooled batches range from 50 – 70 donors, but large batches may have up to 200. While some degree of variability from lot to lot exists, each lot is required to meet published test specifications. Does this product contain Estradiol or progesterone or is is already stripped

This product is not charcoal-stripped or hormone-depleted. As a human serum product derived from pooled male plasma from blood group AB, this product may contain naturally occurring levels of estradiol and progesterone. Concentrations of estradiol and progesterone are not listed as tested specifications and are therefore not provided.

Is this product obtained from healthy individuals? I noted that each donor unit used in the preparation was tested and found negative for HBsAG, HIV and etc. However, does each donor was tested for any underlying diseases like Diabetes and medically fit?

This serum product is pooled from multiple donors. Typical pooled batches range from 50 – 70 donors, but large batches may have up to 200. The general health and/or underlying condition information is not collected from individual donors. Based on electrophoresis studies, the pooled final product may be considered ‘Normal’ serum.

Is this serum contain Uric acid? If yes how much, or how to check it?. If the serum is expired, is it okay to use?

While uric acid is present in human serum, this product is not tested for uric acid levels. Should uric acid measurement is required, see product MAK483, Uric Acid Assay Kit, which is suitable for the determination of Uric acid in serum. It is not recommended to use products that are beyond the assigned expiration date.

Once the product has been thawed, how should it be stored? How many times can be freezed and thawed?
