Pyrophosphatase aus E. coli (E-PPase) besitzt einen breiteren optimalen pH-Bereich und enthält vier divalente Kationen pro Untereinheit. Sie enthält 175 Aminosäuren mit zusätzlichen Aspartatresten im Aktivstellenhohlraum. Strukturell ist E-PPase ein Homohexamer mit sechs identischen 20-kDa-Untereinheiten. Magnesium ist ein Cofaktor für E-PPase.
