Anwendung

BME ist geeignet für die Reduzierung von Protein-Disulfidbindungen vor der Polyacrylamidgel-Elektrophorese und ist für gewöhnlich in einer Konzentration von 5 % in einem Probenpuffer für SDS-PAGE enthalten. Die Aufspaltung intermolekularer Disulfidbindungen (zwischen Untereinheiten) ermöglicht, dass sich die Untereinheiten eines Proteins bei der SDS-PAGE eigenständig voneinander trennen. Die Aufspaltung intramolekularer Disulfidbindungen (innerhalb von Untereinheiten) sorgt dafür, dass die Untereinheiten komplett denaturiert werden, sodass die einzelnen Peptide gemäß ihrer Kettenlänge und nicht in Abhängigkeit ihrer Sekundärstruktur migrieren.
Reduktionsmittel
β-Mercaptoethanol oder 2-Mercaptoethanol wurde verwendet:
  • als Zusatz im Roswell Park Memorial Institute (RPMI)-1640 Kulturmedium für die Züchtung von DT40-Zellen (Hühner-B-Zelllinie)[1]
  • für die Lyse von NuLi-1-Zellen (Normale Lunge, University of Iowa-1)[2]
  • für die Trypsinisierung von embryonalen Stammzellen der Maus[3]

  1. In which buffer or solvent should 2-Mercaptoethanol be diluted for working solution preparation?

    1. 2-Mercaptoethanol is compatible with a wide range of common aqueous buffers, such as Laemmli sample buffer, phosphate-buffered saline, or Tris buffer. Certain buffers, such as Tris, can cause oxidation of 2-mercaptoethanol. In these situations, preparation of the solution is recommended just before use.

  2. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.

    1. This product is a neat or pure material. The concentration is considered to be 100%. The calculated molarity is 14.3 M as reported in the Properties section of the product page.
      To prepare a 50 mM solution, please mix 0.391 g of M3148 with 100 mL solvent. 0.391 g / 78.13 g/mol / 0.1 L = 0.05 = 50 mM.
