General description
Subtilisin ist eine nicht-glycosylierte Einzelpolypeptidkette ohne Disulfidbindungen und besitzt ein Molekulargewicht von 27 KDa.
Application
Dieses Produkt ist ein proteolytisches Enzym, das aus der Fermentation von Bacillus licheniformis isoliert wird. Es ist eine Serin-Endoproteinase mit einer breiten Spezifität für native und denaturierte Proteine, die unter alkalischen Bedingungen aktiv ist. Produkt P8038, auch als Subtilisin Carlsberg bekannt, wurde zur Hydrolyse von Herzzellen für die Untersuchung der Stilllegung von kardialen mitochondrialen NHE1 eingesetzt.
Das Enzym von Sigma wurde zur Optimierung der Freisetzung aller mitochondrialen Populationen aus homogenisiertem Ventrikelgewebe von Rattenherzen eingesetzt. Es wurde außerdem bei der Vorhybridisierungsbehandlung formalinfixierter Paraffinschnitte von Leberproben zum Nachweis humaner und viraler DNA verwendet.
Biochem/physiol Actions
Proteinase katabolisiert Proteine durch Hydrolyse von Peptidbindungen. Proteasen werden durch Serin-Aktivstelleninhibitoren wie Phenylmethylsulfonylfluorid (PMSF) und Diisopropylfluorphosphat inaktiviert.
Subtilisin A gehört zur Familie der Serin S8-Endoproteinasen. Es weist eine breite Spezifität mit Präferenz für ein großen ungeladenen Rest an Position P1 auf. Es hydrolysiert native und denaturierte Proteine und ist unter alkalischen Bedingungen aktiv.

Eine Einheit hydrolysiert die Caseinmenge, die eine Färbung entsprechend 1,0 μmol (181 μg) Tyrosin pro min bei pH 7,5 und 37 °C produziert (Farbe vom Folin-Ciocalteu Reagenz).
