Beschreibung

Application

Eine thermostabile (thermophile) extrazelluläre Metallendopeptidase mit vier Calciumionen. Kofaktoren sind Zink und Calcium. Hydrolysiert Proteinbindungen an der N-terminalen Seite von hydrophoben Aminosäureresten. Der optimale pH-Wert beträgt 8,0 und die optimale Temperatur für Aktivität beträgt 70 °C. Beachtliche Stabilität bei einem pH-Wert von 5 bis 9,5. Thermolysin weist eine geringe Spaltungsspezifizität auf, weshalb es eine Reihe von kurzen Fragmenten erzeugt, die sich zur Sequenzierung eignen. Bevorzugte Spaltung: X-Spaltung-Y-Z, wobei X = eine beliebige Aminosäure; Y = Leu, Phe, Ile, Val, Met, Ala und Z = eine beliebige Aminosäure außer Pro. Eine Spaltung am N-Terminus von Leu wird gegenüber einer Spaltung am N-Terminus von Phe bevorzugt, die gegenüber den anderen bevorzugt wird. Wird häufig verwendet, um eine eingeschränkte Proteolyse für das Peptid-Mapping und Untersuchungen der Proteinstruktur und konformationellen Veränderungen durchzuführen.
