Descrizione

General description

Tris-Borate-EDTA (TBE) buffer is a commonly used running buffer for nucleic gel electrophoresis with a pH range from 8.1-8.5 (25 °C). TBE buffer has a higher buffer capacity than TAE buffer and has a higher resolution for DNA fragments <1500 bp. Borate in TBE buffer acts as an enzyme inhibitor.

Application

Il tampone di migrazione TBE è il tampone più usato nell′elettroforesi su gel di poliacrilammide per DNA ed RNA. Il TBE si usa con gel non denaturanti e denaturanti (urea 7 M). È usato abitualmente anche per il gel di sequenziamento automatizzato del DNA. Il TBE può essere usato anche per il gel di agarosio ma è sconsigliato con i gel preparatori per il recupero degli acidi nucleici.
La diluizione dei concentrati madre di TBE fino a un tampone di migrazione TBE 1x dà luogo a un tampone che contiene 89 mM di Tris-borato e 2 mM di EDTA, a pH 8,3. Le scorte 5x o 10x si possono anche aggiungere a una soluzione madre di acrilammide/bis-acrilammide per realizzare il gel PAGE. Per ottenere la massima risoluzione si consiglia l′applicazione di tensioni inferiori a 5 V/cm (distanza tra gli elettrodi dell′unità).

Suitable as a running and gel preparation buffer in:

• DNA agarose gel electrophoresis (fragment <1500 bp)
• Native and non-denaturing RNA agarose gel electrophoresis (fragment <1500 bp)
• Denaturing gradient gel electrophoresis
• Northern and Southern blotting
• DNA automated sequencing gel
• Pulsed-field gel electrophoresis (PFGE) (applied voltages of less than 5 V/cm are recommended for maximum resolution)

Preparation Note

Preparato con base Trizma Biotechnology Performance Certified (con certificazione della prestazione biotecnologica) (codice prodotto T6066) e acido borico (codice prodotto B6768) ed EDTA sale disodico (codice prodotto E5134) come reagenti per biologia molecolare.

Contents of one pouch, when dissolved in 500mL of distilled or deionized water, will yield a 1X solution containing 89mM Tris-Borate, 2mM EDTA, pH 8.3 at 25 °C. Certified to be DNAse, RNAse, Protease, and Nickase free. Certified for use in electrophoresis. Recommended temperature for dissolution 5 °C to 40 °C.

Store at room temperature. Product may naturally agglomerate but can be simply broken up within the pouch prior to use.
