General description

L-lysine is a homopolypeptide having two enantiomeric forms, L and D lysine. Through its three reactive groups (the carboxylic acid and the α and ε amines), L-lysine has the capacity to generate various branched polymeric structures, including dendrimers, branched polylysine, and hyperbranched polylysine (PLL). [1] Poly-L-lysine is a polymer made up of positively charged amino acid residues, with each lysine residue typically containing one HBr.

Application

Los polímeros de poli-L-lisina pueden utilizarse en la promoción de la adhesión celular a los sustratos sólidos, la conjugación al metotrexato para incrementar el transporte de medicamentos, la microencapsulación de islote, la tecnología de microencapsulación celular, recubrimiento de portaobjetos de vidrio de micromatrices y preparaciones cromosómicas. La poli-L-lisina de menor peso molecular (30 000-70 000) es menos viscosa en disolución, pero las versiones de mayor peso molecular proporcionan mas sitios de unión por molécula.
Poly-L-lysine has been used:

  • In coating 96-well plates for cell attachment in testing respiratory burst activity.[2]
  • In coating glass cover slips for immunofluorescent labeling of intracellular structures.[3]
  • For pretreating 6-well plate in myosatellite cell isolation protocol.[4]
  • In coating coverslips for cell adhesion in E14 neuron culture to study possible mechanism of action of circRNA_01477 in neurons.[5]

Biochem/physiol Actions

La poli-L-lisina es un factor de unión inespecífica a las células útil en la promoción de la adhesión celular a los sustratos sólidos intensificando la interacción electrostática entre iones con carga negativa de la membrana celular y la superficie del cultivo. Cuando se absorbe a la superficie del cultivo celular, la poli-L-lisina funciona aumentando el número de sitios con carga positiva disponibles para la unión a la célula. Con las células que pueden digerir la poli-L-lisina, debe utilizarse la poli-D-lisina como factor de fijación.
Poly‐l‐lysine (PLL) acts as polycationic polymers that have been extensively used to carry drugs, genes etc. across the cell membrane. [6] Poly-L-lysine (PLL) has the capability to engage in the formation of polyelectrolyte complexes (PECs) by interacting with anionic entities such as polyanions, nucleic acid chains and some drugs.[6]

Preparation Note

This product is supplied as a 0.01% sterile filtered solution in water, using a molecular weight of 150 - 300 kDa. Coating slides with this solution should be followed with a 5 minute incubation and drying the slides at room temperature. Coated slides will be stable for one year if protected from dust.

La poli-L-lisina es una polímero de aminoácido con carga positiva con aproximadamente un HBr por residuo de lisina. El hidrobromuro permite a la poli-L-lisina estar en una forma cristalina hidrosoluble. Puede encontrarse una pequeña cantidad del producto en la estructura beta porque el HBr interfiere en el enlace de hidrógeno entre el grupo amino y los grupos carboxilo o los grupos que contienen N u O.

    1. Yes, this product is suitable for coating culture surfaces such as plates, flasks, slides, and cover slips. After the coated surface is completely dry store in a sealed container or ziplock for up to one year. Do not use if coated surface becomes yellow or cloudy.
