Application

Hyaluronidase from bovine testes has been used for the digestion of breast tissue, heart tissue and oocytes for isolation of cells.[1][2][3]

Suitable for use in preparation of single cell suspension for sequencing.

Biochem/physiol Actions

Hyaluronidase degrades hyaluronan and has been found to be inappropriately regulated during cancer progression.
These enzymes randomly cleave β-N-acetylhexosamine-[1→4] glycosidic bonds in hyaluronic acid, chondroitin, and chondroitin sulfates.

Other Notes

One unit will degrade 0.75 μg of the polysaccharide hyaluronic acid per minute at pH 5.35 at 37 ºC (as measured by turbidimetric absorbance (λ: 600nm) when complexed with BSA after 45 minutes).

Protocols

To measure hyaluronidase activity, a turbidimetric determination assay is used at 600 nm. One unit of hyaluronidase activity will cause a change in absorbance of 0.330 per minute at pH 5.35 at 37 °C.

    1. This product is NOT tested for injection into animals. The product is NOT sterile or tested for endotoxin levels.

  2. Is there any information available regarding whether hyaluronidase impacts body fluid crystals?

    1. Hyaluronidase is not expected to impact crystals. It is specific to hyaluronan or Hyaluronic acid and may be used with breast and possibly other tissues. However, crystals are typically inorganic salts, with body fluid crystals typically being pyrophosphate. Therefore, hyaluronidase should have no effect upon pyrophosphate crystals.

  3. Is it acceptable to use H3506 SLCH0779 directly in synovial fluid without dilution?

    1. It is generally not a standard practice to simply sprinkle hyaluronidase (such as H3506 SLCH0779) directly into synovial fluid. For a more formal and generally recognized method, a 0.05% solution in phosphate buffer is recommended, with one drop added per milliliter of fluid. This allows for the clarification of synovial fluids to make cell counts possible. Therefore, directly using H3506 SLCH0779 in synovial fluid without dilution may not be the most common practice.

    1. The type designations for the hyaluronidase enzyme products offered are in-house designations and do not represent any kind of biochemical significance. The types can refer to the order of addition of the product to the catalog, source, physical form, activity, tests performed, etc.

  5. Can I dissolve H3506 in DMEM/F12 media or PBS before use for tissue dissociation?

    1. It is recommended to dissolve this product at 1 mg/mL in 0.02 M phosphate buffer, pH 7, containing 77 mM sodium chloride and 0.01% BSA. Solubilizing directly into the medium has not been tested, but may be suitable. It is important to include BSA to ensure stability.

  7. How does the storage temperature relate to shipping conditions?

    1. The storage conditions that a Sigma-Aldrich catalog and label recommend for products are deliberately conservative. For many products, long-term storage at low temperatures will increase the time during which they are expected to remain in specification and therefore are labeled accordingly. Where short-term storage, shipping time frame, or exposure to conditions other than those recommended for long-term storage will not affect product quality, Sigma-Aldrich will ship at ambient temperature. The products sensitive to short-term exposure to conditions other than their recommended long-term storage are shipped on wet or dry ice. Ambient temperature shipping helps to control shipping costs for our customers. At any time, our customers can request wet- or dry-ice shipment, but the special handling is at customer expense if our product history indicates that the product is stable for regular shipment.
