Application

Asialofetuin from fetal calf serum is suitable for use:
  • in the assay of the lactose binding activity of the galectin-7 by immunodetection of in situ binding activity to asialofetuin immobilized on nitrocellulose[1]
  • in the inhibition of zona pellucida hardening of mouse oocytes[2]
  • as an acceptor in sialyltransferase enzyme activity assay[3]
Incorporation of asialofetuin (AF) in liposomes strongly enhance delivery to and endocytosis by cells displaying the AF receptor, notably hepatocytes. This provides a very efficient route for gene therapy.

Biochem/physiol Actions

Feutins are glycoproteins that belong to the cystatin superfamily. They are characterized by the presence of two tandemly arranged cystatin-like domains and an unrelated domain.[4] It binds to transforming growth factor (TGF)-β/BMP (bone morphogenic protein) cytokines.[5]
Asialofetuin (AF) binds to asialoglycoprotein receptor (ASGP-R) and is employed as a ligand for drug delivery to the hepatocytes. It aids serum-resistant lipoplexes for efficient gene deliver into the HepG2 (human hepatoblastoma) cells.

    1. The difference between the Type I and Type II Asialofetuin products does not represent any kind of biochemical or functional difference. These indicate different methods of production and purification. The Type II option, product A1908, utilizes neuraminadase in an enzymatic cleavage step. The Type I, product A4781, utilizes a proprietary modification of that described in Spiro, R.G., J. Biol. Chem., 235, 2860 (1960).
