General description

Recombinant human PDGF‑BB is a homodimeric growth factor expressed in E. coli and supplied as a lyophilized powder for mammalian cell culture research applications in vitro.

Application

Platelet-Derived Growth Factor-BB human has been used :

• As a growth supplement in proximal tubule and glomerular explant cell culture
• In three-dimensional scaffold preparation
• To study the effect of brivanib on growth factor signaling pathways
• In transwell migration assay

Biochem/physiol Actions

Platelet-derived growth factor (PDGF) elicits multifunctional actions with a variety of cells. It is mitogenic to mesoderm-derived cells, such as dermal and tendon fibroblasts, vascular smooth muscle cells, glial cells, and chondrocytes. PDGF significantly activates chemotaxis of neutrophils, monocytes, and fibroblasts. It increases the synthesis of phospholipids, cholesterol esters, glycogen and prostaglandins, and modulates LDL (low density lipoprotein) receptor binding.
PDGF stimulates fibroblast proliferation and extracellular matrix production in order to facilitate tissue repair. PDGF-BB isomer is found to be critical in angiogenesis and is highly expressed in tumor, mediating oncogenic pericyte recruitment. PDGF-BB is an important regulator of bone homeostasis, repair and regeneration.

Physical form

Lyophilized from a 0.2 μm filtered solution in 30 mM acetic acid.

Analysis Note

The biological activity is measured by its ability to stimulate ³H-thymidine incorporation in quiescent NR6R-3T3 fibroblasts.
