General description
The ZytoLight SPEC FGFR3 Dual Color Break Apart Probe (PL124) is a direct-labeled FISH probe designed for the qualitative detection of translocations involving the human FGFR3 gene at 4p16.3 in formalin-fixed, paraffin-embedded (FFPE) specimens. It is intended for use in conjunction with the ZytoLight FISH-Tissue Implementation Kit.

Fluorescence in situ hybridization (FISH) uses direct-labeled probes flanking FGFR3 breakpoint that co-denature with target DNA in pretreated FFPE sections (10 µl/slide, sealed), hybridize overnight (75°C/10 min denaturation, 37°C hybridization), then undergo stringent washing and DAPI counterstaining for visualization via fluorescence microscopy with color-specific filters per ZytoLight FISH-Tissue Implementation Kit.
Components:
ZytoLight®SPEC FGFR3 Dual Color Break Apart Probe comprises:
- ZyGreen labeled polynucleotides (excitation 503 nm/emission 528 nm) targeting the FGFR3 gene region on chromosome 4 (4p16.3).
- ZyOrange labeled polynucleotides (excitation 547 nm/emission 572 nm) targeting sequences proximal to the FGFR3 breakpoint region.
- Formamide-based hybridization buffer.

Analysis Note
In a normal interphase nucleus, two green signals (for FGFR3) and two orange signals (for the corresponding breakpoint) are expected. An aberrant signal pattern may consist of multiple green signals, indicating the presence of a translocation. Any unexpected signal patterns should be further investigated.
