Biochem/physiol Actions

胰蛋白酶是一种丝氨酸内肽酶。在 pH 7.5-9 时,特异性水解赖氨酸和精氨酸的蛋白和肽键 C 端。Arg 和 Lys 的酰胺和酯键也发生断裂。以胰岛素(胰岛素 B ox )的氧化 B 链作为底物,验证了胰蛋白酶测序级(修饰)的特异性。将高浓度的胰蛋白酶(测序级)(经修饰,一份按重量酶加 9 份按重量胰岛素 B ox )孵育 18 小时,以检测痕量糜蛋白酶杂质。

General description

从牛胰腺中分离得到的测序级胰蛋白酶(经过修饰的)是一种高度纯化的特异性蛋白酶,随后进行修饰。

胰蛋白酶是一种高效的特异性蛋白酶,被广泛用于蛋白质组学中的蛋白消化。它产生具有独特特征的短肽,可兼容当前的分离和鉴定方法[6] 比如液相色谱、质谱(MS)。[7][8]

抑制剂:
TLCK、DFP、PMSF、亮肽素、大豆胰蛋白酶抑制剂、鸡蛋来源的胰蛋白酶抑制剂、抑肽酶、 α2-巨球蛋白,α1-抗胰蛋白酶、APMSF 和止痛剂。

保存条件(工作液):-15~-25℃
胰蛋白酶测序级,经过改造,即使在 pH 值处于中性和弱碱性范围时,也较耐自溶。该酶可以高浓度使用。
2-8°C 条件下储存时,1% 乙酸或 1 mM hcl 溶液的稳定期限为 1 周。-15--25°C 条件下储存时,该溶液至少可稳定 1 年而不损失活性。
