测序级胰蛋白酶的参数是通过氧化的胰岛素B-链 （胰岛素Box）作为底物而被确认的。高浓度的测序级胰蛋白酶（1份重量的酶及18份重要的胰岛素Box）孵育18小时后以检测胰凝乳蛋白酶杂质的存在。
参数（HPC，与胰岛素Box）：
1小时后断裂：≥90%;
18小时后非特异性断裂：≤10%;

Preparation Note
稳定剂：胰蛋白酶在4 M是稳定的。
工作浓度：蛋白重要的1/100至1/20（溶液中）；1-5 μg/100 μl （用于胶内消化）
储存条件（工作溶液）：在0.01% (v/v)三氟乙酸或1 mM HCl中的溶液，如果储存在2到8°C，可最多使用一周。在中性至微碱性pH值的溶液中蛋白的孵育可能会发生部分的自溶。对于此应用，Roche推荐测序级的改良胰蛋白酶。
