Keep using our User Guide for guidance. A 3 kda membrane is extremely small and every 3 minutes counts in having water pass through the membrane. One would want to continue spinning at that speed or shortly below. In time, the initial volume does decrease. One cannot concentrate water. A purpose of using water initially is simply to wet the membrane before introducing a real sample in the hopes of decreasing adherence of the membrane to the substance to be concentrated. One can simply spin at 500 x g for a few moments then discard, then introduce the sample.

General description
El diseño vertical y la superficie de membrana disponible proporcionan un procesamiento rápido de la muestra, una elevada recuperación de la muestra (normalmente superior al 90 % de la disolución inicial diluida) y la capacidad de concentración de 30 veces. La polarización del soluto y el consiguiente ensuciamiento de la membrana se reducen al mínimo gracias al diseño vertical, y un bloqueo físico del filtro evita la centrifugación hasta la sequedad y la posible pérdida de muestra. La recuperación eficiente de la muestra concentrada (especies retenidas) se consigue mediante una cómoda etapa de espín inverso después de recoger el filtrado.

I would like to use this device to remove protein from human serum. Afterwards I would need to use the filtered serum in cell culture conditions. Is there also a sterile version of this product available? Or can this product safely be autoclaved?

Amicon Ultra devices are used to concentrate substances in liquids. For example, if a protein of interest is 68 kDa, a device with a 30 kDa molecular weight cutoff would be used to concentrate it. The 68 kDa protein would remain on top of the filter and not pass through, while smaller substances might be present in both the filtrate and the retentate. These devices concentrate rather than purify. A sterile version of this product is not available, but a method to sanitize involves adding 70% isopropyl alcohol, spinning for a minute, and then discarding. This product cannot be autoclaved.

What is the margin of error for the MWCO? I read somewhere the RECOMMENDATION is to use a MWCO 2-3x smaller than your target. But I was wondering what’s the standard deviation from 3 kDa?

For most membranes, the nominal molecular weight limit, also referred to as the molecular weight cut-off, is determined experimentally under a standard set of operating conditions. These analyses typically employ purified globular proteins to serve as markers or indicators of the retention characteristics of an ultrafiltration membrane. An ultrafiltration membrane with a stated nominal molecular weight limit should retain (reject) at least 90% of a globular solute of that molecular weight in daltons. However, for a wider safety margin, the selected cut-off should be well below the molecular weight of the solute to be retained. It is recommended to use a cut-off 2-3 times smaller than the molecular weight of the protein to be concentrated.

Is this product compatible with organic solvents, such as DMSO or DMF?

The Amicon Ultra products are suitable for use with samples containing up to 10% DMSO and 10% DMF. Exposure to 100% DMSO is not recommended. Unfortunately, the compatibility of these devices with 100% DMF has not been determined.

Is the 0.5 ml 3KDa Amicon device (UFC5003) compatible with a solution containing 5% SDS (Sodium Dodecyl Sulfate)?

The Amicon® Ultra centrifugal devices are designed for processing biological fluids and aqueous solutions. Amicon® Ultra Filter Devices are compatible with concentrations of Sodium dodecyl sulfate (SDS)of 0.1% or less. Prior to use, it is important to verify the chemical compatibility of the sample with the device.

How long should I centrifugate at 14.000 g and how long for the reverse spin at 1000 g?

Typical concentration time for the 3kD membrane device is approximately 30 minutes. However, spin times may need to be adjusted to either a shorter or longer time depending on the sample starting volume, initial concentration, composition, and desired minimum volume.
