Application

Concanavalin A from Canavalia ensiformis (jack bean) has been used:

• to induce spleen blastogenesis
• to generate ConA blast targets from B6 and lpr spleen cells
• to stimulate splenic lymphocytes from estrogen-and placebo-treated mice

Biochem/physiol Actions

Con A ist nicht blutgruppenspezifisch, hat aber eine Affinität für terminale α-D-Mannosyl- und α-D-Glucosylreste. Für ihre Aktivität sind Ca²⁺- und Mn²⁺-Ione erforderlich. Bei einem pH-Wert von maximal 5,6 löst sich Con A in Dimere auf. Bei einem pH-Wert von 5,8 bis 7,0 liegt Con A als Tetramer vor; bei einem pH-Wert über 7,0 bilden sich Aggregate. Con A weist eine mitogene Aktivität auf, die vom Ausmaß der Aggregation abhängt. Succinylierung führt zu einer aktiven dimeren Form, welche bei einem pH-Wert von über 5,6 ihre Form beibehält.

Where reported, agglutination activity is expressed in μg/ml and is determined from serial dilutions in phosphate buffered saline, pH 6.8, containing Ca²⁺ and Mn²⁺ of a 1 mg per mL solution. This activity is the lowest concentration to agglutinate a 2% suspension of human erythrocytes after 1 hr incubation at 25 °C.
