General description

Das Glutathion-S-Transferase(GST)-Assaykit wurde zur Messung der Gesamt-GST-Aktivität entwickelt. Es kann zum Messen der GST-Aktivität in Zell- und Bakterienlysaten, Gewebehomogenaten und Plasma-
und Erythrozytenlysaten eingesetzt werden.

Application

Das Glutathion-S-Transferase-Assaykit nutzt 1-Chlor-2,4-dinitrobenzol (CDNB), das sich für das breiteste Spektrum von GST-Isozymen eignet. Bei der Konjugation der Thiolgruppe von Glutathion an das CDNB-Substrat kommt es zu einer Steigerung der Absorption bei 340 nm.

Biochem/physiol Actions

Glutathion-S-Transferasen (GSTs) sind eine Gruppe von Enzymen, die bei der Entgiftung zahlreicher Xenobiotika in Säugern wichtig sind. Die Enzyme schützen die Zellen vor Giftstoffen, indem sie die Thiolgruppe des Glutathions an elektrophile Xenobiotika konjugieren und die Zellen auf diese Weise vor den mutagenen, karzinogenen und toxischen Wirkungen der Verbindungen schützen. Die GST-Aktivität tritt in Pflanzen, Insekten, Hefen, Bakterien und den meisten Säugergeweben auf, besonders aber in der Leber, der bei der Entgiftung eine wichtige Funktion zufällt.

    1. Is this kit suitable to measure GST enzyme activity in plants? If yes, would plant tissue homogenate will work for this kit?

      1. This kit is not tested specifically for use in plant tissue. However this kit is based on the same principal as referenced in Mozer, T. J., et al., Purification and characterization of corn glutathione S-transferase. Biochemistry 22, 1068-1072 (1983) and is expected to be suitable. This kit has also been used successfully to measure GST activity in plants by other researcher. Please see the following link to review one such example:

    2. How can I determine the shelf life / expiration / retest date of this product?

      1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
        For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
        For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

    3. How is shipping temperature determined? And how is it related to the product storage temperature?

      1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature.     4. Is there a protocol available for preparing tissue for assay with the CS0410 Glutathione-S-Transferase (GST) Assay Kit?

      1. A specific tissue preparation protocol is not included with this kit. Product C3228, CelLytic MT reagent for mammalian tissue, is suitable. Please see the link below to review the product datasheet and protocol:

    5. Is this suitable for detecting all of GST activity or is there for specific GST family for example GSTA or GSTM activity only?

      1. This product has not been evaluated for specificity to each GST isozyme. However, there is no evidence to suggest that this kit does not react to all GST isozymes.
