General description
Mitomycin C ist eine systemische Chemotherapieverbindung und ein Antibiotikum, das aus Streptomyces caespitosus gewonnen wird. Es ist das erste bioreduktive Alkylierungsmittel, das identifiziert wurde, und ist ein Derivat von 3-Amino-5-hydroxybenzoesäure (AHBA), D-Glucosamin, L-Methionin und Carbamoylphosphat.

Application
Mitomycin C aus Streptomyces caespitosus wurde für die Behandlung von Feeder-Schichten wie z. B. PMEFs (primäre embryonale Fibroblasten der Maus) und CD1 embryonale Fibroblasten (MEFs) der Maus für die Kultur von humanen embryonalen Stammzellen (hESCs) verwendet. Es wurde außerdem für die für die Behandlung von BLC-Zelllinien (Basal-like-Karzinome) eingesetzt.
Biochem/physiol Actions
Mitomycin C bewirkt die Vernetzung doppelsträngiger DNA, was zu Mutagenese, Hemmung der DNA-Synthese, Auslösung der DNA-Reparatur und Aktivierung der Apoptose führt. Die Fähigkeit, die DNA-Synthese zu hemmen, ist durch die in Mitomycin C vorhandene Aminochinongruppe bedingt. Diese Verbindung hat keine Auswirkungen auf die Synthese von RNA und Protein. In Gewebekulturen verringert diese Verbindung die Lebensfähigkeit der Zellen, unterdrückt die Mitose und verursacht eine Desorganisation des Zellkerns und die Produktion von Riesenzellen. Mitomycin C besitzt starke anti-gramnegative und anti-grampositive Aktivitäten. Es enthält drei Gruppen: die Chinin-, die Urethan- und die Aziridingruppe. Es wird verwendet, um mitotisch inaktive Feeder-Zellen in Zellkultursystemen zu erzeugen, z. B. die mitotisch inaktiven Fibroblasten, die in Embryonalstammzellsystemen verwendet werden.
Wirkungsmechanismus: Dieses Produkt ist ein Alkylierungsmittel, das spezifisch auf die Guaninnukleosidsequenz 5′-CpG-3′ abzielt. Es hemmt die DNA-Synthese, indem es kovalent mit der DNA reagiert und Vernetzungen zwischen komplementären DNA-Strängen bildet. Diese Wechselwirkung verhindert die Trennung der komplementären DNA-Stränge und hemmt somit die DNA-Replikation.
Antimikrobielles Spektrum: Mitomycin C weist eine starke Antitumorwirkung, insbesondere gegen Ehrlich-Aszites-Tumorzellen, sowie eine starke bakterizide Wirkung gegen grampositive und gramnegative Bakterien auf.

Preparation Note
Mitomycin C ist bei 0,5 mg/ml wasserlöslich, mit einem pH-Wert von 6,0 bis 7,5. In sauren Lösungen mit einem pH-Wert <6 wird es einer schnellen Zersetzung unterzogen, und behält seine Wirkung am wahrscheinlichsten in Lösungen mit einem pH-Wert von 6 bis 9 bei.
Disclaimer
Dieses Fläschchen enthält 2 mg Mitomycin C und 48 mg NaCl. Stammlösungen sollten mittels Filtration sterilisiert und bei 2 bis 8 °C im Dunkeln gelagert werden. Lösungen mit einem pH-Wert von 6 bis 9 können bei 0 bis 5 °C bis zu einer Woche gelagert werden, wenn sich aber eine Ausfällung bildet, sollte eine frische Lösung hergestellt werden. Die ausgefällte Lösung erwies sich als toxisch gegenüber Zellen.
