Structural maintenance of chromosomes protein 1A (UniProt: Q14683; also known as SMC protein 1A, SMC-1-alpha, SMC-1A, Sb1.8) is encoded by the SMC1A (also known as DXS423E, KIAA0178, SB1.8, SMC1, SMC1L1) gene (Gene ID: 8243) in human. SMC1 is a ATPase and subunit of the multiprotein cohesion complex, a protein involved in chromosome segregation and in double-strand DNA break (DSB) repair. The cohesion multiprotein complex is required for sister chromatid cohesion. This complex is composed partly of two structural maintenance of chromosomes (SMC) proteins, SMC3 and either SMC1B or the protein encoded by this gene. Most of the cohesion complexes dissociate from the chromosomes before mitosis, although those complexes at the kinetochore remain. Therefore, the encoded protein is thought to be an important part of functional kinetochores. Structurally, SMC1A consists of five distinct regions: two coiled-coil domains, a hinge domain, and N- and C-terminal domains, which include ATP and DNA binding motifs. It is ubiquitously expressed, with higher levels found in lymph nodes, appendix, and colon. SMC1A is predominantly localized in the nucleus. The protein also interacts with BRCA1 and ATM proteins indicating a role in DNA repair. ATM phosphorylates SMC1 at Ser957 and Ser966 both in vitro and in vivo. SMC1A at Ser966 is also phosphorylated by ATR kinases. Phosphorylated SMC1 is part of the complex that functions as a downstream effector in both the ATM/NBS1 branch and the ATR/MSH2 branch of S-phase checkpoint. Mutations in this gene result in Cornelia de Lange syndrome. It is reportedly involved in the G2/M transition of human glioma cells and G1/S transition of human lung adenocarcinoma cells. However, recent studies have also suggested its pivotal role in epithelial mesenchymal transition, tumorigenesis, metastasis, and chemo- and radio-resistance in cancer cells. Immunogen

KLH-conjugated linear peptide corresponding to 11 amino acids surrounding phosphoserine 966 from the C-terminal half of human SMC1A.

Anwendung

Quality Control Testing

Evaluated by Western Blotting in HeLa cells treated with Etoposide.

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected SMC1A phosphorylated at serine 966 in lysate from HeLa cells treated with Etoposide (20 g/mL; 3 h) but, not in untreated HeLa cell lysate.

Tested Applications

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected SMC1A phosphorylated at Ser966 in HeLa cells treated with Etoposide (20 g/mL for 3 hours, post serum starvation overnight) but, not in untreated HeLa cells.

Peptide Inhibition Assay: Target band detection in lysate from HeLa cells treated with Etoposide (20 g/mL; 3 h) was prevented by pre-blocking of a representative lot with the immunogen phospho-peptide, but not with the corresponding non-phospho peptide.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Clone 1B11 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects SMC1A phosphorylated at serine 966.

Physikalische Form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Angaben zur Herstellung

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
