Descrizione generale

L′insulina è un ormone polipeptidico a doppia catena prodotto dalle cellule β delle isole pancreatiche. Il suo peso molecolare è circa 5800 Da. Le catene α e β sono unite da due legami disolfuro. La catena α contiene un legame disolfuro interno.

Applicazioni

La soluzione di insulina umana è raccomandata per applicazioni di coltura cellulare a concentrazioni tra 0,5 e 1mL per litro di terreno.
Una soluzione di insulina umana è stata usata per test di tolleranza all′insulina in topi e ratti.[1][2] È anche stata usata per la differenziazione adipogenica di colture derivate da cellule staminali embrionali umane (hESCs) e di hMSCs (cellule stromali mesenchimali umane).[3][4]
La soluzione di insulina umana è stata utilizzata come supplemento al terreno Eagle modificato da Dulbecco (DMEM) per la coltura di una linea di cellule epiteliali e di una linea cellulare endometriale umana, al terreno essenziale minimo di Eagle per la coltura di una linea di cellule di adenocarcinoma mammario umano e al terreno 199 per la coltura di una linea cellulare ovaria normale.

Azioni biochim/fisiol

L′insulina è coinvolta in vari processi cellulari, dalla promozione della crescita delle cellule, alla regolazione del ciclo cellulare, fino alla promozione dell′assimilazione del glucosio. Viene usato normalmente come supplemento proteico nelle colture di cellule di mammifero prive di siero per contribuire a stimolare la crescita e il mantenimento più adeguati alle cellule mammifere.

    1. This product is recommended to be stored at 2-8°C. For research applications, freezing insulin solution is a common practice. However, solutions available on the commercial market for therapeutic use are not frozen to maintain maximum activity. No stability studies have been performed on the shelf life of frozen insulin solutions.
