Application

Bovine serum albumin has been used-
  • as a component of PCR (polymerase chain reaction) mix
  • in the preparation of dissociated cortical neurons
  • during immunofluorescent labeling
  • as a component of Ca2+-free saline (containing collagenase, hyaluronidase and DNAse) used for bovine tissue digestion

Biochem/physiol Actions

Certain conformational and primary-sequence epitopes of BSA are suspected allergens in human beef and milk allergies.

Preparation Note

Prepared using heat shock fractionation.
Serum albumin may be referred to as Fraction V. This naming convention is taken from the original Cohn method of fractionating serum proteins using cold ethanol precipitation. Serum albumin was found in the fifth ethanol fraction using Cohn′s method. Since then, the term "Fraction V" has been used by some to describe serum albumin regardless of the method of preparation. Others have used this term to describe serum albumin purified by ethanol fractionation methods that have been highly modified since the original Cohn method was described. Sigma-Aldrich manufactures and distributes serum albumins purified from a variety of primary methods including the true Cohn fractionation method, modified ethanol fractionation methods, heat shock and chromatography. Additional purification steps may include crystallization or charcoal filtration.

    1. This product has not been validated for cell culture use on batch release. It has been primarily cited for immunoassay applications, with a few references to cell culture applications. Please see the links below to BSA products tested for suitability in cell culture:

  3. How long is BSA (A7906) viable at room temperature? 200g was left on benchtop overnight (Approximately 22 hours), will it still work?

    1. This item is stored refrigerated at 2-8°C. However, the product is shipped at ambient temperature. Shipping routes are configured to minimize transit time.
      Refrigerated products that ship at ambient temperature are considered stable up to 2 weeks for transit time.

  5. After reconstitution in PBS, for how long is the protein stable in solution if stored at 4°C or -20*C?

    1. Prepared BSA solution products also available for purchase. These products have a shelf life ranging from 18 months to 2 years. Many are assigned a restest schedule with no firm expiration date. These products are prepared in saline, DPBS, or Tris. These solutions are filtered products and offered in a range of concentrations. The storage recommendation for these products is 2 - 8 deg. C. In addition, BSA is often used as a carrier protein for less stable compounds that are stored frozen. While there is no established shelf life for a BSA solution stored frozen, it is quite stable.   6. Does this BSA have bovine immunoglobulins?

    1. This product does contain bovine immunoglobulins. BSA products that do not contain immunoglobulins will state "globulin-free" or "essentially globulin-free" on the product detail page. Product A3156 is an example of an immunoglobulin free BSA product.

  7. Can Product A7906, Albumin from bovine serum (BSA), be used in immunohistochemistry?

    1. We have not ourselves tested A7906 for use in immunohistochemistry. However, a literature search can turn up publications that cite this use, such as this paper: Surendran, K., et al., Wnt-Dependent Beta-Catenin Signaling Is Activated after Unilateral Ureteral Obstruction, and Recombinant Secreted Frizzled-Related Protein 4 Alters the Progression of Renal Fibrosis. J. Am. Soc. Nephrol., Vol. 16, pp. 2373-2384 (2005).

    1. We have not tested A7906 in quality control to be protease-free. Other BSA products to suggest that are protease-free would be A3059 or B6917.

    1. We have not tested the use of A7906 in ELISA. Because the remainder of the product is mostly globulins, we would not suggest A7906 for ELISA use.
