Amicon® Ultra 15 mL离心装置与Ultracel® 10,000 Dalton分子量截止滤波器提供快速超滤,以及可靠的大分子保留。例如,0.25 mg/ml初始浓度下的细胞色素C (12.4 kDa)保留率为>95%。

    1. The spin conditions for a swinging-bucker rotor are 4000 x g and for a fixed-angle rotor are 5000 x g, at a time range of 10 - 60 minutes, based on molecular weight cut-off.

  2. Is there a recommended temperature for the amicon ultra-15? I would like to concentrate protein at 0 degrees C. Also, is glycerol able to pass through the 30kD filter? The enzyme I want to concentrate is in 50% glycerol. Thanks!

    1. No data is available on the maximum or minimum temperature for using the Amicon Ultra-15. Results shown in the user guides are based on experiments conducted at approximately 25°C, not with refrigerated centrifuges. Some users work at 4°C, but filtration times are longer. No studies have been performed at 0°C. Glycerol is compatible with this product.

  3. Can I concentrate cell-propagated respiratory syncytial virus stock using the Amicon® Ultra Centrifugal Filter, 10 kDa MWCO? what is your though and suggestion?

    1. Concentrating cell-propagated respiratory syncytial virus stock with this product has not been studied. However, it can be attempted, as other viruses such as SARS, AAV, and Lenti have been successfully concentrated. The product concentrates substances larger than the molecular weight cut-off of the membrane. It is recommended to consult online research articles using keywords like "Amicon and respiratory syncytial virus" on platforms such as PubMed or Google Scholar.

  4. Do you make a miniature version of the Amicon® Ultra Centrifugal Filter for 1.5ml samples?

    1. ​Yes, ultrafiltration of protein samples can potentially lead to lower migration on an SDS-PAGE gel.During the ultrafiltration process, the desalting or buffer exchange step may remove or reduce sodium chloride (NaCl), leading to lower protein migration on the gel. To prepare protein samples for gel electrophoresis, using an Amicon Ultra (AU) device or a suitable desalting/buffer exchange method is recommended.

  7. Hello, I'm using UFC903096 (Amicon-15, 30K) and am trying to exchange and concentrate without over-concentrating <500uL ... Would you have a table of the volume to add to the filtrate collector to ensure ~ final product volumes. Thanks!

  8. How to wash the column after one time used for the buffer exchange or for protein concentration. Is any protocol please share??

    1. We do not recommend reusing the device because eventually the membrane will clog completely. We have yet to record an efficient way to clean the pores of the membrane without damaging the membrane itself.
