一般說明

3-异丁基-1-甲基黄嘌呤/IBMX是cAMP和cGMP磷酸二酯酶的非选择性、非特异性抑制剂。[1]IBMX可诱导黑色素生成,并可以用作黑色素生成研究的阳性对照。[2] 在卵母细胞研究中,IBMX有助于维持前期I卵母细胞的生发泡期(GV)阻滞。

應用

3-异丁基-1-甲基黄嘌呤已用于:
  • 分化间充质干细胞 (MSC)[3]
  • 用作诱导脂肪形成的培养基补充剂[4]
  • 用作Krebs-Ringer缓冲液和HEPES (KRBH)溶液的补充剂,诱导葡萄糖刺激的胰岛素分泌(GSIS)[5]
  • 鉴定脂肪来源的间充质干细胞[6]
  • 环磷酸腺苷(cAMP)测定[7]
  • 磷酸二酯酶(PDE)抑制[8]

生化/生理作用

由IBMX对磷酸二酯酶的抑制而引起的cAMP水平的增加可激活PKA,而引起增殖减少、分化增加并诱导细胞凋亡。IBMX可抑制苯肾上腺素诱导的气道粘膜神经内分泌上皮细胞5-羟色胺释放。(IC50:1.3 μM)。IBMX还可作为一种腺苷受体拮抗剂。其已表现出对神经肌肉接头、GH3细胞和血管平滑肌细胞中离子通道的抑制作用。IBMX可诱导感觉神经元释放储存在细胞内的钙。

  2. After dissolving 3-Isobutyl-1-méthylxanthine (I5879) in DMSO or in pre-warmed methanol, where should it be stored (long term or short term)?

    1. Solutions in DMSO may be stored at -20 °C. The stability of this material in methanol has not been investigated. However, an external publication reports storage of IBMX solutions in ethanol at 0.125 mg/mL at 4 °C for at least three months.     1. This material is soluble at a maximum concentration of 1M in DMSO. The 200 mM concentration is cited in Plouffe, B. et al., Methods Enzymol., 484, 295-328 (2010).
      This concentration is sufficient for most applications. The product is tested for solubility in pre-warmed methanol at 50 mg/mL. The preferred solvent and stock concentration is dependent upon the application and requirements of the end user.
