Gene Information

human ... FN1(2335)

General description

Fibronectin 1 is a glycoprotein of the extracellular matrix that is coded by FN1 gene. It is expressed in the plasma and at the cell surface. The FN1 gene mapped to human chromosome 2q35.
FN occurs as a dimeric protein, linked by two disulfide bonds at the end of C-terminal. Within the fibronectin structure, the domains are compactly arranged close to both the amino and carboxyl terminal of the peptide chains. Moreover, it has been observed that despite arising from a single gene up to 20 distinct variants of this gene have been identified in humans.

Application

Fibronectin from human plasma has been used in the determination of H2O2 consumption rate.[1] It has also been used in coating the Sylgard® models for cell culture. It has also been used in cell adhesion and to directly pattern the protein onto the polymeric film.

Biochem/physiol Actions

Fibronectin function encompasses a diverse range of biological processes including cell adhesion, growth, migration, wound healing, blood coagulation and metastasis. Fibronectin protein plays an important role in cell attachment and spreading, control of cell cytoskeleton, morphology and differentiation. FN1 is also involved in extracellular matrix formation, hemostasis and thrombosis. Mutations in FN1 results in glomerulopathy.[2]

Features and Benefits

  • Adhesive glycoprotein
  • Derived from human plasma
  • Recommended surface coverage of 1-5 μg/cm2

Preparation Note

This product is a 0.1% solution of fibronectin. The protein homogeneity is evaluated by immunoelectrophoresis, is endotoxin tested, and the source material was tested negative for HIV, hepatitis B and hepatitis C.

In coating culture surfaces, fibronectin should be diluted in sterile balanced sterile salt solution and coated with minimal volume. Surface should then be air dried for 45 minutes at room temp and can be stored for 2-4 weeks at 2-8°C

Analysis Note

Protein homogeneity is evaluated by immunoelectrophoresis.

Questions

1–9 of 9 Questions
  1. Could Fibronectin (F0895) be used as a positive control for fibronectin in western blot assay? If so, do one need to consider any extra steps in the preparation of such ctrl sample?

    1 answer
    1. This product is intended for use as an attachment substrate for cell culture and has not been tested in western blotting as a positive control. However, the purification process of the starting material includes an affinity step and immunoelectrophoresis performed during Quality testing is against Anti-Fibronectin antibody F3648. While the end user would have to determine suitability, this material is expected to be suitable for use as a positive control in western blotting.

      Helpful?

  2. How many µg/cm² would the coating concentration be if I applied a coating with a 0.1% concentration using the F0895 product?

    1 answer
    1. The concentration of the 0.1% solution can be found in the Certificate of Analysis. To calculate micrograms per square centimeter, consider the volume used and the surface area of the well.

      Helpful?

  3. What is the recommended dilution rate for coating the culture surface with F0895-1MG Fibronectin as per the preparation instructions?

    1 answer
    1. The recommended dilution rate for coating the culture surface with F0895-1MG Fibronectin, as per the preparation instructions, is to start with a concentration in the range of 1 - 5 ug/cm^2. For a 96-well plate well with a surface area of about 0.34 cm^2, approximately 2 ul of F0895 further diluted in saline with a total volume of around 50 ul would be sufficient to cover the bottom of the well.

      Helpful?

  4. What is the recommended storage duration for Fibronectin (F0895) at 4°C, -20°C, and -80°C? How long can a diluted solution of 10ug/mL in PBS be stored at 4°C? Is it possible to reuse the Fibronectin solution to coat culture ware surfaces multiple times?

    1 answer
    1. The recommended storage duration for Fibronectin (F0895) is up to 1 year at 4°C. There has been no testing done on its stability after storage at -80°C or -20°C. A diluted solution of 10ug/mL in PBS can be stored at 4°C for 2-4 weeks. However, the solution cannot be reused to coat culture ware surfaces multiple times; the recommended process involves coating surfaces with a minimal volume, allowing them to air dry for 45 minutes at room temperature, and then storing the cultureware at 4°C for 2-4 weeks.

      Helpful?

  5. Is Product F0895, Fibronectin from human plasma, sterile?

    1 answer
    1. Fibronectin 0.1% solution is a sterile product.  It is tested for sterility.  The sterility test results can be found on the lot-specific certificate of analysis.

    1. Transportation information can be found in Section 14 of the product's (M)SDS.To access the shipping information for this material, use the link on the product detail page for the product.

      Helpful?

  7. How should I store and handle Product F0895, Fibronectin from human plasma?

    1 answer
    1. Recommended storage of this material is  2-8 °C.  Vortexing, excessive agitation, repeated freezing and thawing of reconstituted fibronectin are not recommended.

      Helpful?

  8. What is the solvent for Product F0895, Fibronectin from human plasma?

    1 answer

Why do you not have a protocol for using this expensive reagent?

You have no protocol online for using this very expensive item......why? I have been waiting to chat for a total of 15 minutes to get what you should have put online anyway

No issues, seems to work for hiPSC differentiation protocols.

Used multiple times and different lots for hiPSC differentiation into iMeLCs. No problems with it.
