General description

Serum-free culture environment avoids the variability and contaminants that can be introduced by serum. Serum-free formulations that substitute a purified form of the factors normally supplied by serum are suitable for many in vitro growth and differentiation studies. These factors include insulin, transferrin, selenium, pyruvate, and ethanolamine. The addition of other components varies greatly, depending on the cell type being studied and the basal medium employed. ITS is a mixture of insulin, human transferrin (partially iron-saturated), and sodium selenite. It is a general cell supplement designed for use in non-complex media (e.g., MEM, RPMI-1640) and complex media (e.g., Ham′s F-12, DME/F-12, MEM) with sodium pyruvate.

Application

ITS+1 Liquid Media Supplement (100×) has been used as a supplement in high-glucose Dulbecco′s Modified Eagle Medium (DMEM) for differentiation of mouse mesenchymal stem cells (mMSCs) to chondrocytes. It has also been used as a supplement in basal differentiation medium for differentiation of human-induced pluripotent stem cell (hiPSC) into osteoblasts under serum-free conditions.
Insulin-transferrin-sodium selenite, linoleic-BSA has been used for serum-free cell culture applications.ITS+1 Liquid Media Supplement (100×) has been used in culturing cell lines[1] and for the preparation of immortalisation medium for the production of cell lines from ′Immortomouse′ embryos.[2]

Preparation Note

Prepared in Earle′s Balanced Salt Solution (EBSS) without phenol red.

Other Notes

Contains 1.0 mg/ml bovine insulin, 0.55 mg/ml human transferrin (substantially iron-free), 0.5 μg/ml sodium selenite, 50 mg/ml bovine serum albumin and 470 μg/ml linoleic acid at the 100x concentration.

  1. Could you provide information on the unit of activation for the ITS+1 liquid media supplement - product I2521? The media I want to use requires 0.023U/mL of insulin in 500mL of RPMI, and I need to understand the activation units for the insulin supplement. The product information only mentions a 1mg/mL concentration of insulin.

    1. The activity of the product should be 25 IU/mL, based on the formulation and the IU of the raw materials used. The product has a specification of >= 0.9mg/mL, and the raw material has an activity specification of >= 25 IUM. Therefore, depending on the lot the customer received, if the assay of the lot they received was 1mg/mL, then the activity would be 25 IU/mL.
