The MILLIPLEX® MAP 7-plex DNA Damage/Genotoxicity Magnetic Bead Kit (Cat. No. 48-621MAG) is a premixed kit to be used for the simultaneous quantification of the following 7 analytes in cell lysates:• ATR (total)• Chk1 (Ser345)• Chk2 (Thr68)• H2A.X (Ser139)• MDM2 (total)• p21 (total)• p53 (Ser15)

應用

Intracellular Bead-Based Multiplex Assays using the Luminex technology enables the simultaneous relative quantitation of multiple phosphorylation and total pathway proteins in tissue and cell lysate samples. Compare Multiplexing results to those of Western blotting.An overnight (4°C) incubation is recommended for best results.This assay requires 25 μL diluted cell lysate per well.This kit must be run using Assay Buffer 1 (provided).The suggested working range of protein concentration for the assay is 1 to 25 μg of total protein/well (25 μL/well at 40 to 1,000 μg/mL).Analytes available:ATR (Total);Chk1 (Ser345);Chk2 (Thr68);H2A.X (Ser139);MDM2 (total);p21 (Total);p53 (Ser15)

Questions

  1. Is it possible to incorporate GAPDH as a loading control into the DNA Damage panel (48-621MAG) even though it specifies the use of Assay Buffer 1 and the GAPDH MAPmate (46-667MAG) requires Assay Buffer 2? Does the difference in assay buffer formulations affect the compatibility of adding GAPDH to the panel?

    1. GAPDH (46-667MAG) is functional only in Assay Buffer 1 when used at significantly lower dilutions, approximately 1 microgram of total protein per well. This specificity is the reason it is designated for use with Buffer 2 only. Most panels necessitate higher protein concentrations, making it impractical to multiplex GAPDH with panels that utilize Assay Buffer 1. When conducting assays with Assay Buffer 1, B-Tubulin MAPMate (46-713MAG) is the recommended alternative.
