Our laboratory has not specifically studied the impact of high salt, such as 500 mM NaCl, on the activity of RNAseA. However, Park and Raines (2003) have investigated the activity of RNAseA under high salt concentration, up to 1 M NaCl. The results available in the publication titled "Catalysis by ribonuclease A is limited by the rate of substrate association" may serve as a useful reference. The study is available in Biochemistry, 2003 Apr 1;42(12):3509-18. From the study, it is noted that "the pKa values at 1.0 M NaCl are expected to be higher by 0.4-0.5 unit than the pKa values determined at 0.142 M NaCl."
