Application
ヒト血漿由来アポリポタンパク質A-Iは、ニトロベンゾオキサジアゾール（NBD）-コレルテロール担持細胞においてコレステロール流出を誘発するために使用されています。また、酵素結合免疫吸着測定法（ELISA）を用いてIgG抗アポA-1抗体を検出するためにも使用されています。
高密度リポタンパク質(HDL)の主要なタンパク質成分です。
Biochem/physiol Actions
Apo A-IはHDLのタンパク部分の約70%を占めています。Apo A-Iはアミノ酸残基数245個の1本鎖ポリペプチドで、C末端にグルタミン酸を、N末端にアスパラギン酸を有します。分子量は28.3 kDaと報告されています。Apo A-Iは、1種類のメジャ-なイソ型(pI 5.6)と2種類のマイナ-なイソ型(pI 5.53および5.46)からなります。Apo A-Iは、αらせん構造を多く含んでいます。αらせん構造に存在する両親媒性領域は、脂質結合能に関与していると考えられています。水溶液中では、apo A-Iは自己会合し、わずかに構造変化します。Apo A-Iは、血漿中のコレステロ-ルのエステル化に関与するlecithin-cholesterol acyltransferase(LCAT)を活性化します。
Apo A-Iの血漿濃度の正常値は90~130 mg/dlです。Apo A-I値は、冠動脈疾患のリスクと負の相関を示す可能性があります。
