General description

Endothelin-1 (ET-1) is synthesized and secreted by activated endothelial cells, and is a peptide with potent vasoconstrictor activity. It is a marker for activated or dysfunctional endothelial cells.[1] This peptide is one of the three isopeptides belonging to the endothelin family. It is also released by cardiomyocytes.[2]

Application

Endothelin-1 (ET-1) has been used to study the production and detection of H2O2 and production of NO in the presence of ET-1,[3] for studying the effects of ET-1 on the eNOS/inducible NOS (iNOS)/caveolin-1 expression, NO production, isoform specific PKC translocation, and PKC activity in human saphenous vein endothelial cells (HSVECs).[4]

Biochem/physiol Actions

Endothelin is a potent agonist for ETA and ETB endothelin receptors.
Endothelin-1 (ET-1) is a vasoconstrictor, which is increased in plasma and explants of patients with uterine leiomyomas. Thus, uterine fibroids are linked to increased ET-1 levels and hypertension, which is worsened by hypoxic condtions.[1] ET-1 RNAi and Endostar are capable of preventing the progression and invasiveness of PC-3 (prostate cancer) cells.[5] The plasma levels of this peptide are increased in patients with atrial septal defect (ASD), and these levels might help in diagnosis of closure defects.[2] The plasma level of ET-1 is also higher in patients with end-stage renal disease (ESRD), and this is linked to gradual worsening of pulmonary function tests.[6]
Potent agonist for ETA and ETB endothelin receptors. Induces the production of hypoxia-inducible factor 1α, and, thus, the production of VEGF.
