Apostatin-1 is a cell penetrant apoptosis inhibitor that activates autophagy leading to restoration of cellular homeostasis in cells with accumulated mutant tau, α-synuclein, or huntingtin. Apostatin-1 potently and selectively inhibits TRADD. It binds to the N-terminal TRAF2-binding domain of TRADD (TRADD-N). Apparently Apostatin-1 releases cIAP1/2 or TRAF2 from TRADD thus promoting the K63-linked ubiquitination of beclin 1.

A cell-permeable derivative of podophyllotoxin that acts as a topoisomerase II inhibitor (IC50 = 59.2 µM) has major activity against a number of tumors, including germ cell neoplasms, small cell lung cancer, and malignant lymphoma.
