Gene Information
human ... CIROP(100128908)
General description
Ciliated left-right organizer metallopeptidase (UniProt: A0A1B0GTW7; also known as EC:3.4.24.-, Leishmanolysin-like peptidase 2) is encoded by the CIROP (also known as LMLN2) gene (Gene ID: 100128908) in human. The ciliated left-right organizer metallopeptidase (CIROP) is a zinc-dependent metalloprotease crucial for establishing left-right asymmetry during vertebrate embryonic development. Encoded by the CIROP gene, this protein features a signal peptide, a zinc catalytic domain with a consensus active site motif (His-Glu-x-x-His), a cysteine-rich domain, and a C-terminal transmembrane domain. CIROP is specifically expressed in ciliated left-right organizers (LROs), such as the node in mice and Kupffer′s vesicle in zebrafish, where it is essential for proper function of motile cilia. These cilia generate directional fluid flow that triggers asymmetric activation of the Nodal pathway, a signaling cascade responsible for the left-right organization of internal organs. In terms of subcellular localization, CIROP is membrane-associated, with its catalytic domain exposed extracellularly. It interacts with other proteins and substrates localized in the LRO to regulate signaling and structural components of the cilia. Dysregulation of CIROP or mutations in its gene are implicated in heterotaxy syndrome (HTX12), an autosomal recessive disorder characterized by randomization of organ placement along the left-right axis, often accompanied by severe congenital malformations, especially in the cardiovascular system. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Application
Evaluated by Western Blotting in CIROP-transfected HEK-293T cell lysate.
Western Blotting Analysis: A 1:40,000 dilution of this antibody detected CIROP in CIROP-transfected HEK-293T cell lysate.
Tested Applications
Western Blotting Analysis: A 1:40,000 dilution from a representative lot detected CIROP in CIROP-transfected HEK-293T cell lysate, but not in untransfected HEK-293T cell lysate.
Affinity Binding Assay: A representative lot of this antibody bound with CIROP peptide with at least one million-fold (1,000,000X) higher affinity than with non-specific control peptide.
Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected CIROP in HEK-293T cells transfected with CIROP-Strep-Tag.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
