Applicazioni

La streptomicina è usata per studiare il meccanismo di resistenza nei suoi confronti e spesso è impiegata insieme alla penicillina e ad altri agenti per inibire la contaminazione batterica in applicazioni con colture cellulari. Nelle applicazioni di biologia molecolare se ne raccomanda l′uso a 25-50 μg/mL, nelle colture cellulari a 100 mg/L e nella coltura di embrioni a 50 mg/L.

Azioni biochim/fisiol

Modalità d′azione: La streptomicina agisce inibendo la sintesi delle proteine dei procarioti legandosi alla proteina S12 della subunità ribosomiale 30S. In questo modo impedisce la transizione da complesso imitativo a ribosoma di allungamento della catena. Ciò causa errori di codifica o inibisce l′avvio.

Modalità di resistenza: Una mutazione in rpsL, un gene per la proteina ribosomiale S12, impedisce il legame della streptomicina con il ribosoma. Inoltre, un′aminoglicoside fosfotransferasi inattiva la streptomicina.

Spettro antimicrobico: La streptomicina è attiva nei confronti dei batteri gram positivi e gram negativi.

Nota sulla preparazione

Le soluzioni madre devono essere preparate direttamente nel flaconcino con concentrazioni di acqua sterile tra 20 e 25 mg/mL.

Tenere il contenitore ben chiuso in luogo asciutto e ben ventilato. Conservare in luogo asciutto.

Esclusione di responsabilità

Le soluzioni possono essere conservate a 2-8 °C per non più di un mese oppure a -20 °C per periodi più prolungati. A 37 °C le soluzioni sono stabili per tre giorni.
