Application
このキットには、真菌および細菌増殖培地、マクロファージライセートおよび精製酵素調製物中のキチナーゼ活性を効率的に検出するために必要なすべての試薬が含まれています。さらに、このキットには、さまざまな種類のキチン分解活性の検出に使用できる次の3通りの基質が含まれています。
- 4-ニトロフェニルN,N′-ジアセチル-β-D-キトビオシド。エキソキチナーゼ活性（キトビオシダーゼ活性）に適した基質。
- 4-ニトロフェニルN-アセチル-β-D-グルコサミニド。エキソキチナーゼ活性（β-N-アセチルグルコサミニダーゼ活性）の検出に適した基質。
Biochem/physiol Actions
また、キチン分解酵素は、キチン基質に対する酵素作用に基づいても分類されます。エンドキチナーゼは、キチン鎖の内部点でのランダムな切断を触媒する酵素として定義されています。エキソキチナーゼは、キチンの非還元末端からアセチルキトビオースまたはN-アセチルグルコサミンを順次切り出していくのを触媒し、それぞれキトビオシダーゼ、β-N-アセチルグルコサミニダーゼと呼ばれます。
キチナーゼは、さまざまな生物でさまざまな機能を果たします。細菌では、主に栄養プロセスに関与しています。酵母やさまざまな真菌では、形態形成に関与しています。動物や植物では、キチナーゼは主に病原体の攻撃に対する防御で役割を果たします。

How is shipping temperature determined? And how is it related to the product storage temperature?

Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers. Hello, I purchasee chitinase assay kit CS0980 last month. Can I use the kiy to analyse chitinase in hepatopancreas of lobsters? I dont find the procedure in the kit to prepare and measure chitinase from tissue samples

As noted in its Technical Bulletin, the kit has been tested on Trichoderma viride, Streptomyces griseus and human macrophages. No protocol is available for the use of the kit to determine Chitinase activity in hepatopancreas of lobsters.

How do you make the substrate in Product CS0980, Chitinase Assay Kit, go into solution?

The substrates do not dissolve easily in the buffer. It may take ~1 hour of shaking to completely dissolve the substrates. Use of a larger container (15 mL) may aid dissolution.

What is the recommended substrate concentration for Product CS0980, Chitinase Assay Kit?

A 1mg/mL substrate solution is recommended. If required, the 4-Nitrophenyl-beta-D-N,N',N''-triacetylchitotriose and the 4-Nitrophenyl N,N'-diacetyl-beta-D-chitobioside can be used at lower concentrations in the reaction (0.5 or 0.2mg/mL respectively).
