酵母ヘキソキナーゼには2種類の互いに類似するアイソフォーム、PI(A)とPII(B)があり、等電点はそれぞれ5.25と4です。

至適pH:7.5～9.0
吸光係数:E1% = 8.85(PI)、9.47(PII)(280 nm)
活性化因子:ヘキソキナーゼは活性にMg2+イオンを必要とします(KM = 2.6 mM)。 ヘキソキナーゼはカテコールアミンとその類縁物質により活性化されます。
阻害剤:ソルボース1-リン酸、ポリリン酸、6-デオキシ-6-フルオログルコース、2-C-ヒドロキシ-メチルグルコース、キシロース、リクソース、チオール反応性化合物(Hg2+および4-クロロ第二水銀安息香酸)
