General description

GenEluteエンドトキシンフリープラスミドキットは、高効率トランスフェクションを実現するために、≤0.1 EU/μg DNAのプラスミドDNAを精製するための、簡便、迅速、かつ費用対効果の高い方法を提供します。 エンドトキシン（リポ多糖類またはLPSとも呼ばれます）はプラスミドDNAと共精製されることが多く、エンドトキシン感受性細胞株ではトランスフェクション効率を著しく低下させます。



組換え大腸菌は、一晩培養したものから遠心分離により回収され、改良アルカリSDS溶解法にかけることで清澄化されたライセートが得られます。清澄化されたライセートからエンドトキシンは、簡単な抽出-相分離ステップで除去されます。プラスミドDNAは、高濃度塩存在下でシリカに吸着させることによりさらに精製されます。簡単なスピンウォッシュステップの後、結合したプラスミドDNAはエンドトキシンフリー水で溶出されます。25～200 mLの培養物から、最大1.2 mgのエンドトキシンフリーのプラスミドDNAを調製できます。

回収されたプラスミドDNAは主にスーパーコイル型です。DNAは、トランスフェクション、制限消化、クローニング、シーケンシング、PCR増幅などの下流アプリケーションですぐに使用できます。
