一般說明

一种细胞可渗透的喹啉基-氨基苯并噻唑化合物，据报告可作为RIP3选择性激酶抑制剂，其选择性是包括RIP1在内的300多种其他激酶的绝大多数的>1,000倍。RIP1抑制剂Nec-1（30 µM；目录号480065 & 505224）和基因敲除可与GSK′872（3µM）结合使用，以阐释在存在或不存在胱天蛋白酶抑制剂Z-VAD-FMK（25µ M；目录号627609 & 627610）的情况下，Toll样受体（TLR）介导的坏死和TNFR1引发的坏死性凋亡中的RIP1依赖性与RIP3依赖性。虽然显示RIP3及其下游底物MLKL在鼠巨噬细胞（BMDM& J774），成纤维细胞（3T3-SA &MEF）和内皮（SVEC4-10）培养物中在存在Z-VAD-FMK时介导TLR3引发的坏死，仅在BMDM& J774巨噬细胞培养物中，RIP1参与也见于TLR3介导的坏死。
