一般說明

MTT是一种淡黄底物,可被活细胞裂解产生深蓝色甲臜产物。这个过程需要活跃的线粒体,即使是新死亡的细胞也不会裂解大量的MTT。下文所述的比色测定法可用于增殖或补体介导的细胞毒性测定。

應用

程序:
1)使用标准方法,在光学质量良好的96孔平底组织培养板中进行淋巴因子、有丝分裂原或补体介导的细胞毒性试验(例如Falcon)。每孔中组织培养基的最终体积应为0.1 mL,培养基(例如RPMI或DMEM)可能含有高达10%的胎牛血清。

2)在试验结束时,向每个孔中加入0.01 mL AB溶液(MTT)。轻轻敲击托盘侧面进行混合。

3)在37°C下孵育,使MTT发生裂解。最佳时间可能因含量测定而异,但4小时适用于大多数目的。在这个时间结束时,在含有活细胞的孔中产生的MTT甲臜会在孔底部显示为黑色的、模糊晶体。

4)向每个孔中加入含0.04 N HCl的0.1 mL异丙醇。用多通道移液器反复移液,充分混匀。HCl将组织培养基中的酚红转化为不干扰MTT甲臜测量的黄色。异丙醇溶解甲臜,得到适合吸光度测量的均匀蓝色溶液。

5)一小时内,在ELISA酶标仪上测定吸光度,测试波长为570 nm,参考波长为630 nm。在室温下放置数小时后,血清蛋白可能会由于酸/酒精而开始沉淀。冷却盘子会加速沉淀。如果测定前必须储存平板,在加入酸/酒精前保持在4°C,然后在读数前加热至室温并加入酸/酒精。

结果:
MTT测定法通常可检测典型细胞系的200至50,000个细胞,尽管1,000至50,000是有用的范围。对于其他细胞类型,此数字可能会有所不同。应建立细胞毒性测定,以使对照,未裂解的细胞的信号为0.2至0.4,并且增殖测定应在平台浓度下产生相似的值。对于典型的细胞系,这相当于每孔约20-50,000个细胞。

吸光度与细胞数成正比;即使浓度高达1 x 106个细胞/mL,实际细胞也不会显著吸收。

準備報告

可在2-8°C保存长达6个月。试剂A/溶液B混合物在2-8°C下可稳定储存长达两周。

试剂制备:
将10 mL溶液B加入一瓶试剂A中。混匀,无菌过滤,并在4°C下避光保存直至使用。注意:可能需要一夜才能溶解。请勿加热溶液。当绝对需要溶解晶体时,用1-2滴HCl调节pH值。在这些条件下,AB混合物可稳定数周。

  1. What is the difference between TOX1 and CT02 ?

    1. These kits are very comparable in terms of usage, and are identical in pricipal. Both kits utilize MTT to measure cell expansion, The CT02 kit is suitable for 1000 tests when using a 96 well formt. The TOX1 recommends adding reagents as a percentage of the culture volume to allow for alternative formats. In addition, the CT02 kit allows for the use of phenol red in culture medium, whereas the TOX1 kit recommends the use of phenol red free medium. The wavelength at which the test is read does vary slightly. These kits have not been tested in a side by side comparison. Please see the links below to review the protocol for each product.

  2. Can the mixture of solution A and B be stored at -20ºC for a long term use?

    1. As mentioned in the protocol listed in the DESCRIPTION section under 'Procedure', the Reagent A / Solution B mixture is stable at 2-8°C for up to two weeks. The freezing is not recommended as it is more likely to fall out of the solution upon freezing.
