General description
Dithiothreitol (DTT), ein Schutzmittel für die Thiolgruppe,[1] ist ein hervorragendes Reagenz, um die Oxidation von SH-Gruppen in Proteinen zu verhindern. Die DTT-Lösung ist ein starkes und vielseitiges Mittel zur Reduzierung von Disulfiden.[2] Es reduziert eine Disulfidbindung, indem es einen Sechsmembranring bildet und einen Thiol-Disulfid-Austausch einleitet.
Application
DL-Dithiothreitol-Lösung wird wie folgt verwendet:
- bei der Behandlung von Darmexplantaten
- bei der Homogenataufbereitung von Kiemenzellen für enzymatische Assays
- bei der Proteinelektrophorese für Western Blotting
- bei der Transglutaminaseaktivität
- im Probenpuffer für SDS-PAGE
- in Western Blots als Bestandteil des Lysepuffers für die Einzelkern-RNA-Sequenzierung
- als Bestandteil von Proteinextraktionspuffer für die Zelllyse
Biochem/physiol Actions
DL-Dithiothreitol (DTT) wird in einem chemischen Debonding-Verfahren eingesetzt, das dabei hilft, Bakterien von Biofilmen zu entfernen. Es kann die strukturelle Stabilität des GABA(B)- (γ-Aminobuttersäure-)Rezeptors verändern, indem es die Disulfidbindungen zwischen vier Cysteinresten, die sich an der GABA(B1(a))-Rezeptorstruktur befinden, inaktiviert.Dithiothreitol (DTT) wird in großem Umfang bei der synthetischen Peptidsynthese und bei der biochemischen Bildung von Thiolproteinen verwendet. Auch in Studien zu verschiedenen Bereichen der Proteinchemie wie Enzymaktivität und Proteinfaltung kann es zum Einsatz kommen. DTT steuert explizit die Thiol-Disulfid-Austauschreaktion, um die intra- oder intermolekularen subatomischen Disulfidbindungen in Biomolekülen vollständig zu reduzieren. Die Produkte dieser Reaktion sind Thiole und das zyklische Disulfid von DTT.

How can I determine the shelf life / expiration / retest date of this product?

If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.

If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature.
