General description
アミロイドβタンパク質断片1-40（Aβ40）は、ヒト染色体21q21.3にマップされる、アミロイドβタンパク質（Aβ）に由来します。Aβ40は主に血管のアミロイド沈着物に存在します。Aβ40はC末端に膜挿入ドメインを有します。水-ミセル媒体において、ランダムコイルからα-ヘリックス構造への構造転移を示します。
Application
アミロイドβタンパク質断片1-40は以下の用途に使用されています：
- フーリエ変換赤外分光法／示差走査熱量測定法（FT-IR/DSC）を用いた、温度に基づくコンホメーションの検討
- 頭部外傷患者の脳脊髄液中のアミロイドA4タンパク質を定量するための、サンドイッチ型酵素免疫測定法の標準物質
- 線維芽細胞におけるアミロイド沈着を抑制するための、胚性幹細胞培地の一成分[1]
Biochem/physiol Actions
アミロイドβタンパク質は、in vivoにおいて、またヒト、ラットの培養神経細胞などin vitroにおいても神経栄養性や神経毒性を発揮します。βアミロイドペプチド(アミノ酸1-42、1-43)は、アルツハイマー病患者の海馬、新皮質、扁桃体に出現する老人斑や神経原線維変化の主たる構成成分です。
アミロイドβタンパク質断片1-40（Aβ40）は、陽イオンベースのイオンチャネルを形成します。
Preparation Note
ストック溶液をカルシウム不含のPBSで1 mg/mLに希釈し、37°Cで4日間インキュベートしてからご使用いただくと、最大の生物活性が得られます。
