Biochem/physiol Actions

SAvPhire™ Monomeric Streptavidin is a variant of the biotin-binding streptavidin, which naturally exists as a tetramer. Many monomeric streptavidin proteins have rapid dissociation kinetics, which prevent their use in stable labeling applications. However, SAvphire™ Monomeric Streptavidin has been engineered to have a dissociation rate comparable to that of tetrameric streptavidin.
The size and monovalency of this product provide advantages over tetrameric streptavidin in applications such as functional cell surface labeling and atomic force spectroscopy. These advantages include:
  • Minimizing distribution or dynamics in the probing of biotinylated targets
  • Reducing risk of aggregation, precipitation, and cross-linking in pre-incubation steps with biotinylated molecules
  • Easier access to desired target

This product can be used in traditional biotin-binding applications, including Western blotting, immunoprecipitation, immunohistochemistry, and flow cytometry.

Preparation Note

Once reconstituted, the solution can be stored at 2-8 °C for at least 1 month. For long term storage, aliquot and store at -20 °C.
This product is lyophilized from PBS, pH 7.4. Reconstitute the vial contents to a 4 mg/mL solution by adding water as follows:

  • 1 mg vial: add 250 μL of water for a 4 mg/mL solution
  • 5 mg vial: add 1.25 mL of water for a 4 mg/mL solution

The reconstituted product can be further diluted in water or PBS as needed.

  1. I'm intending on succinimidyl ester labelling primary amines of this - could either a mutation map / any information about where the lysines are relative to the biotin binding pocket be provided?

    1. The SAvPhire Monomeric Streptavidin has 5 Lysines and 4 are within the last 18 residues on the C-terminus. The specific sequence is considered proprietay. The number of lysines in the biotin-binding pocket has not been determined.

  2. Could you provide the optimal buffer in which this product best binds biotin?

    1. This material is lyophilized from a PBS solution, pH 7.4 . For assay purposes, the vial is reconstituted using Nuclease, Protease, and RNase-Free Water. Alternatively, the product may also be reconstituted in PBS, see product P3813. Please see the link below to review additional information available in the product datasheet:
