Application

Pyridostatin hydrochloride has been used as a G-quadruplex (G4)-stabilizing ligand to check the effect of G4s on fork progression. It has also been used as a G4-stabilizing ligand to study the effect of G4 on apurinic/apyrimidinic endonuclease 1 (APE1) mediated Kirsten rat sarcoma virus (KRAS) expression in pancreatic ductal adenocarcinoma (PDAC) cell lines.

Biochem/physiol Actions

Pyridostatin is a highly selective G-quadruplex (G4) interacting molecule. It retards the growth of human cancer cells by inducing replication/transcription dependent on DNA damage. Pyridostatin stabilizes G-quadruplexes, targeting the proto-oncogene SRC and telomeric G4, inducing DNA damage and cell-cycle arrest.
Stabilizer of G-quadruplexes (G4).

Questions

  1. Hi, I need to make up a known concentration solution of this compound, but you only supply a free base molecular weight. Can you provide me with a full molecular weight for lot #0000374186? Also, what's the storage and water solubility? Thanks in advance

    1 answer
    1. The solubility of this material is tested in DMSO. The solubility in water has not been determined. However, external sources indicate that Pyridostatin hydrochloride is soluble in water at 1 mM. Solutions may be stored up to 1 month at -20°C (store solutions under nitrogen). The HCl content varies from lot to lot and may range from 4.5 to 5.5 mol/mol. The water content may also range from 0.0 - 7.0 mol/mol. This information is reported in the Certificate of Analysis.
