Descrizione generale

L'eparina è un glicosamminoglicano (GAG) appartenente alla famiglia dei mucopolisaccaridi, noto per le sue potenti proprietà anticoagulanti.[1] Questo polisaccaride estensivamente solfatato è composto da D-glucosammina e acido exuronico legati da legami glicosidici.[2] Trova ampia applicazione nella ricerca biochimica e negli studi con colture cellulari.

Applicazioni

Al terreno preparato per l′inseminazione di oociti con spermatozoi è stato aggiunto sale sodico di eparina estratta dalla mucosa intestinale suina.[3][4] Il sale sodico di eparina estratta da mucosa intestinale suina è stato usato:
  • per verificare l'effetto dell'eparina sulla trasduzione del segnale dell'FGF21 (fattore di crescita dei fibroblasti 21);[5][5]
  • per l'immunocolorazione di sezioni cerebrali murine;[6]
  • per quantificare leucociti ed eritrociti in campioni di sangue di Sparus aurata;[7]
  • nel dosaggio dell'anticoagulazione e nei saggi di inibizione del coronavirus 2 responsabile della sindrome respiratoria acuta grave (SARS-CoV-2)[8]

Azioni biochim/fisiol

L′eparina è la principale sostanza responsabile del ritardo nella coagulazione del sangue. Nella cascata della coagulazione potenzia l'inattivazione delle proteasi mediata dall'antitrombina.[9]

L'eparina viene sintetizzata dai mastociti, i quali la rilasciano insieme ad altri mediatori, come l'istamina, che intervengono nelle reazioni allergiche e infiammatorie.[10] Il suo effetto terapeutico si esplica nel contrasto alla trombosi venosa e all'embolia polmonare, nella prevenzione dello sviluppo di nuove trombosi post-trombolisi e nella gestione delle trombosi arteriose in pazienti con infarto acuto del miocardio. L'attività antinfiammatoria dell'eparina interviene in molte condizioni infiammatorie come le ustioni, l'asma bronchiale e la colite ulcerosa.[2]

Altre note

Le soluzioni di eparina non vanno autoclavate perché alle alte temperature può avvenire l'ulteriore formazione di legami crociati tra gli zuccheri. Non va nemmeno congelata.

    1. "Grade I-A" originates from product listings for heparin prior to the implementation of the current catalog numbering system. This older grading system has since been phased out, but the terminology was retained in the descriptions of some current products.     1. This product is a porcine heparin with a molecular weight range of 15-19 kDa. This material is considered a "high molecular weight heparin."
