General description

High quality double-stranded template DNA isolated from the thymus of male and female calves.

Application

Calf thymus DNA is exceptionally useful as a substrate for DNA polymerase assays, in the amplification of very long fragments and as a carrier DNA for precipitations.
Calf thymus DNA is exceptionally useful as a substrate for DNA polymerase assays, in the amplification of very long fragments and as a carrier DNA for precipitations. It was used as a standard to determine DNA content in gels[1] and for DNA quantification by fluorescent assay.[2]

Preparation Note

Dissolve the DNA gently rocking overnight to a final concentration of 1mg/ml in molecular biology grade water.
One mg DNA is approx. 20 A260 units.
Prepared from highly polymerized calf thymus DNA and nicked with DNase using the method of Aposhian and Kornberg.[3]

    1. The molecular weight for D4522 is reported to be between 10-15 million Daltons prior to activation (nicking). 15,000 kilodaltons x (1kb/660 kilodaltons) = 22.72 kb (on average). The average number of base pairs is not measured, and therefore, this calculation is an estimate. We do not have a molecular weight measurement after it's been nicked, but it is expected to be a range below 22.72 kB. We do not have comparison data to inform us of how they may respond differently as a standard curve for a fluorescent assay. We would suggest checking to see if the mechanics behind your assay are length dependent or could be influenced by the ends being nicked or not. Products like D4764 that are deemed stable for long periods of time do not have a noted expiration or retest date.

    1. The reported molecular weight is between 10-15 million Daltons before activation (nicking), primarily as double-stranded DNA. A double-stranded DNA of 1 kb in length has a molecular weight of 660 kiloDalton. A size of 15 million daltons is equivalent to 15,000 kilodaltons. When multiplied by (1kb/660 kilodaltons), it equals 22.72 kb on average. It's important to note that the average number of base pairs is not measured, so this calculation provides an estimate.

Solutions of deoxyribonucleic acid have been stored successfully for several months at 4°C, in 10 mM Tris, pH 7.5 - 8.0, with 1 mM EDTA and without a bacteriostatic agent.  At low concentrations (25 μg/mL), DNA tends to absorb onto the surfaces of plastic tubes.  It is not recommended to store deoxyribonucleic acid in alkaline solutions since it tends to degrade at pH greater than 8.0.

  6. Is BSA (bovine serum albumin) used during the method of preparation for Product D4522, Deoxyribonucleic acid?

    1. During the method of preparation, when deoxyribonucleic acid is nicked, BSA is added to the solution.  This is followed by an extraction method to remove all exogenous protein. We do not, however, claim this deoxyribonucleic acid is 100% clean of all proteins (including BSA).
