General description

Deoxyribonucleic acid−cellulose double-stranded from calf thymus DNA, also known as ctDNA is a natural source of DNA designed for DNA binding agent studies. The detection limits of ctDNA are around 2.0ng and the linear ranges of light scattering intensity (I(LS)) is 0.05-1.5 mµg.[1]

Application

Deoxyribonucleic acid−cellulose double-stranded from calf thymus DNA is used to study the binding parameters and binding specificity of a variety of proteins, nucleotides, and chemical reagents including anti-cancer agents. It is frequently used in pull-down assays and chromatographic procedures.

Analysis Note

May contain a trace of Tris and EDTA.

Questions

1–9 of 9 Questions
  1. Can it be used to evaluate nucleosides from microorganisms? Example: thermonuclease in S. aureus

    1 answer
    1. Unfortunately, this product is not tested for the specific application. A literature search is recommended for further information.

      Helpful?

  2. Do you have recommendations for how to swell the DNA-cellulose and prepare it for subsequent use with DNA-binding proteins?

    1 answer
    1. This product may be swelled in 10 mM tris, 1 mM tetrasodium EDTA buffer (pH 7.9). One gram of DNA-Cellulose will swell to approximately 4 mL of wet resin.

      Helpful?

  3. How to dissolve this product to coat elisa plate, please?

    1 answer
    1. This product is not tested for the suitability with coating ELISA plates. It may be helpful to read through the peer-reviewed articles associated with this product: https://www.sigmaaldrich.com/search/d8515?focus=papers&page=1&perpage=30&sort=relevance&term=d8515&type=citation_search

  4. How do you regenerate the column using Deoxyribonucleic acid-cellulose double-stranded from calf thymus DNA (DNA Cellulose), Product D8515?

    1 answer
    1. A suggested procedure for the regeneration of a DNA-Cellulose column would be to use 5 - 10 column volumes of 2M NaCl in buffer, followed by the same volume of buffer alone.

      Helpful?

  5. Can Product D8515, Deoxyribonucleic acid-cellulose double-stranded from calf thymus DNA, be used for purification of DNA-dependent RNA Polymerase?

    1 answer
    1. We have not tested Product D8515, Deoxyribonucleic acid-cellulose double-stranded from calf thymus DNA for this application. However, the following references utilizes double-stranded DNA cellulose for purification of DNA-dependent RNA Polymerase:J. of Bacteriology, 172, 5624-5630 (1990)Biochem. J.,133, 201-203 (1973)

  6. Can Deoxyribonucleic acid-cellulose double-stranded from calf thymus DNA (DNA Cellulose), Product D8515, be exposed to high salt?

    1 answer
    1. DNA-Cellulose is stable in up to 5M NaCl.  DNA-Cellulose should not be exposed to solutions of NaOH, since NaOH denatures DNA from double-stranded to single-stranded.
