Biochem/physiol Actions
O gel de ECM passa por polimerização ativada termicamente quando levado a 20-40 °C, formando uma membrana basal reconstituída. As células PC12 mostrarão a formação de neurites dentro de 2 dias quando forem cultivadas em uma fina camada desse gel de ECM.

Preparation Note
Este produto é preparado a partir do células Engelbreth-Holm-Swarm de sarcoma de camundongo; dialisado contra clorofórmio. O gel deve ser descongelado durante a noite a 2-8 °C antes do uso e dispensado em uma placa de paredes múltiplas usando uma placa e pipetas pré-resfriadas a 2-8 °C. O gel também pode ser diluído até duas vezes com meio de Eagle modificado por Dulbecco a 2-8 °C, o que deve ser feito antes da adição do gel à placa. O produto se gelifica em 5 minutos a 20 °C. Para manipulações prolongadas, o tratamento deve ser conduzido abaixo de 10 °C. As células podem ser plaqueadas sobre uma fina camada de gel de 0,5 mm ou cultivadas dentro de uma camada de 1 mm. Se as células forem cultivadas dentro da camada, adicione as células ao gel antes do plaqueamento a uma densidade recomendada de 3-4 × 104 células por ml. Para dissociar as células do gel, use protease em PBS sem cálcio, magnésio nem EDTA a uma concentração de 0,6-2,4 unidades por ml.

A MEC foi preparada a uma concentração proteica de 8-12 mg/ml (em DMEM), contendo laminina como componente principal, colágeno tipo IV, proteoglicano de heparan sulfato, entactina e outros componentes secundários. A adição de colágeno tipo IV ao gel de MEC aumenta a polimerização.
O produto contém 50 mg/l de sulfato de gentamicina.

Dispase (Product No. D4818) is a Bacillus-derived neutral protease that is recommended for recovering cells cultured on ECM (Product No. E1270) or Matrigel. Dispase will yield a single cell suspension far more effectively than trypsin, collagenase, or other proteolytic enzymes. Dispase is gentle and will not damage cells harvested for sub-cultivation or other study.
